摘要
采用滤纸片法和打孔法2种抗菌试验方法,考察2-羟基-1-萘醛缩氨基脲(NAHSEM)对金黄色葡萄球菌、大肠杆菌、枯草杆菌的抗菌活性的结果表明,NAHSEM对3种细菌具有较强的抗菌活性。荧光光谱法研究了NAHSEM与牛血清白蛋白(BSA)的结合作用表明,这种结合行为使BSA的内源荧光猝灭。通过猝灭常数、结合常数和结合位点数的计算,证明荧光猝灭机理符合静态机制;NAHSEM和BSA形成1∶1稳定复合物。考察不同温度和酸度下的猝灭作用,进一步证实结合作用主要受范德华力和氢键驱动。
The antibacterial activities of 2-hydroxy-1-naphthalene formaldehyde semicarbazide(NHASEM) against staphylococcus aureus,escherichia coli and bacillus subtilis were studied.The results showed that NHASEM has effective antibacterial activities.The interaction of bovine serum albumin(BSA) with NHASEM was investigated by fluorescence spectroscopy.The results indicated that BSA fluorescence was quenched regularly with the addition of NHASEM.NHASEM could bind with BSA to form a 1∶ 1 complex and the quenching mechanism of BSA affected by NHASEM was a static quenching procedure by calculating of the quenching constant,binding number and binding constant.The fluorescence quenching mechanisms were also studied at different temperature and under different pH.The results indicated that interaction between NHASEM and BSA was driven mainly by the interaction of the hydrogen bonds and van der Waals.
