狭基线纹香茶菜对肝损伤小鼠TNF-α及肝组织超微结构的影响 被引量:1

Effects of extract of Isodon Lophanthoides var.gerardianus (Benth.) Hara on the production of TNF-α and the change of ultrastructure of liver cell in mice induced by concanavalin A

摘要 目的观察狭基线纹香茶菜提取物对免疫性肝损伤小鼠TNF-α释放及肝组织超微结构的影响。方法 50只雄性NIH小鼠,随机分为5组,分别为正常对照组,模型组,狭基线纹香茶菜提取物高、低剂量组(剂量分别为18.2g/kg、4.55g/kg),与环磷酰胺(43mg/kg)对照组。除正常对照组外,其余小鼠于实验首日上午尾静脉注射刀豆蛋白A(ConA)20mg/Kg,并于首日、次日和第3d、第4d下午各给药1次,第5d给药后2h,模型组和各给药组小鼠再次尾静脉注射Con A 20mg/kg,4h后,每组取相同序号小鼠8只,取血检测血清TNF-α活性,每组所剩2只小鼠,8h后处死取肝组织,3%戊二醛固定,切片,染色后,透射电镜观察肝组织超微结构。结果显示模型组TNF-α活性较正常组明显升高,肝组织超微结构发生了显著改变,肝细胞严重肿胀、线粒体破坏、毛细胆管和肝窦腔结构明显变形、中性粒细胞滞留,细胞及细胞器的完整性遭到破坏。狭基线纹香茶菜提取物高、低剂量组TNF-α活性较模型组明显降低(P<0.01、0.05)、肝组织超微结构虽有一定程度改变,但细胞及细胞器的形态及完整性得到保护。结论狭基线纹香茶菜具有较好的保肝作用,其作用机理与抑制TNF-α的产生密切相关。 Objective To investigate effects of extract of Isodon Lophanthoides var.gerardianus(Benth.)Hara(LIVG) on the production of TNF-α and the change of ultrastructure of liver cell in mice induced by concanavalin A.Methods Fifty NIH mice were randomly divided into normal control group,model group,extract groups of LIVG(high-and 1ow-dosage),and cyclophosphamidum group(43mg/kg).In the experimental groups,mice received the extract of ILVG(18.20,4.55g/kg,respectively) by gastric perfusion or injected with cyclophosphamidum into its abdominal cavity daily at the first,the second,the third and fourth day afternoon.Excepts those in normal control group,mice in other groups were injected with Con A(dose of 20mg/Kg) via the tail vein at the first day morning of experiment,On the 5th day,mice were injected with Con A once again with the same dosage 2 hours after the drugs were administered.After 4 hours,the blood of eight mice in each group were obtained and the TNF-α activity was determined.The mice in each group were killed 8 hours later and the change of ultrastructure of the hepatocyte of mice was observede under electron microscope.Results It showed that contents of TNF-α in serum in both groups of LIVG were obviously lower than that of model group(P0.01,0.05),Histopathological change of ultrastructure of hepatocyte were alleviated in both groups of LIVG.Conclusion It suggested that the extract of LIVG possess the effects of protecting liver from immune injury in mice induced by concanavalin A by suppressing the production of TNF-α.
