氧化乐果对雄性小鼠的生殖毒性 被引量:4

Study on Reproductive Toxicity of Omethoate in Male Mice

摘要 目的研究有机磷农药氧化乐果染毒对小鼠睾丸组织琥珀酸脱氢酶(succinic dehydrogenase,SDH)、精子顶体酶活力和精液质量的影响以及胚胎毒性的研究。方法将80只健康清洁级雄性昆明小鼠随机分为1.0、2.0、4.0 mg/kg氧化乐果染毒组和对照(蒸馏水)组,每组20只。采用灌胃方式进行染毒,染毒容量为20 ml/kg,每天1次,连续染毒14 d。染毒结束后,对1/2数量的雄鼠进行精子密度和睾丸组织SDH、精子顶体酶活力的测定;另1/2的雄鼠与雌鼠(按雄∶雌为1∶2)合笼后,进行显性致死试验。结果与对照组比较,2.0、4.0 mg/kg氧化乐果染毒组小鼠精子密度和睾丸组织SDH、精子顶体酶活力均明显降低,差异有统计学意义(P<0.05);且随着氧化乐果染毒剂量的升高,小鼠精子密度和睾丸组织SDH、精子顶体酶活力均呈下降趋势。与对照组比较,4.0 mg/kg氧化乐果染毒组小鼠受孕率以及2.0、4.0 mg/kg氧化乐果染毒组着床数、活胎数、活胎体重明显降低,而死胎数升高,差异均有统计学意义(P<0.05);且随着氧化乐果染毒剂量的升高,小鼠受孕率、着床数、活胎数、活胎体重均呈下降趋势,死胎数呈升高趋势。结论氧化乐果对雄性小鼠生殖系统具有一定的毒性作用。 Objective To explore the adverse effect of omethoate on the activity of succinic dehydrogenase (SDH)in testis,acrosomal enzyme of sperm,the quality of sperm and the embryo toxicity. Methods A total of 80 male KM mice (cleaning grade) were randomly divided into four groups including the dose of 1.0, 2.0 and 4.0 mg/kg omethote and the control group, 20 in each. Omethoate was given through gavage, 20 ml/kg, once a day, for 14 consecutive days. the activities of SDH, acrosomal enzyme of sperm and the number of sperm of 40 male mice were determined. Others were caged together with the female mice to do dominant lethal test. Results Compared with the control group, the number of sperm,the activities of SDH and the acrosomal enzyme of sperm in 2.0 and 4.0 mg/kg omethote group decreased significantly (P〈0.05). And as the omethoate dose increased, the number of sperm, the activity of SDH and the acrosomal enzyme of sperm appeared a downward trend. Compared with the control group, the pregnancy rate in 4.0 mg/kg omethote group, the number of implantation of female mice, the number of live fetus and the weight of live fetus in the 2.0, 4.0 mg/kg omethote group lowered obviously, but the number of stillbirth increased obviously (P〈0.05). And as the omethoate dose was increased, the pregnancy rate, the number of implantation of female mice, the number of live fetus and the weight of live fetus appeared a downward trend, the number of stillbirth presented a upward trend. Conclusion Oral exposure to omethoate has determinate reproductive toxicity to male mice.
