补骨脂水提液对成骨细胞OPG-RANKL mRNA表达的影响 被引量:6

摘要 为了探讨补骨脂水提液对体外培养大鼠成骨细胞核因子-κB受体活化因子配体(RANKL)和骨保护素(OPG)mR-NA表达的影响,选取出生24h内的SD大鼠颅盖骨,依次用胰酶和胶原酶消化分离培养成骨细胞,用不同浓度的补骨脂水提液(10-1,1.0mg/mL和10mg/mL)作用第三代细胞,分别在药物作用24、48h和72h后提取细胞总RNA,用实时荧光定量PCR法检测细胞中RANKL和OPG mRNA。结果显示,补骨脂水提液各浓度均能不同程度地促进成骨细胞OPG mRNA的表达、抑制RANKL mRNA的表达,从而上调OPG/RANKL mRNA比值,其中1.0mg/mL和10mg/mL的补骨脂水提液作用显著。说明补骨脂水提液通过调节成骨细胞分泌OPG和RANKL的量来促进骨形成、抑制骨吸收,从而治疗骨质疏松。 The objective of this study was to investigate the effect of Malaytea Scurfpea Fruit extracts on the expression of RANKL and OPG mRNA in rat osteoblasts. The osteoblasts were derived from calvariae of neonatal rat and digested by trypsin and collagenase. The cultured osteoblasts of the third-generation were exposed to different concentrations of Malaytea Scurfpea Fruit extracts (10-1 , 1.0 and 10mg/ mL). Total RNA was prepared from osteoblasts after 24, 48 and 72 h, respectively, and the expression of RANKL and OPG mRNA was observed by RT-PCR. The results indicated that Malaytea Scurfpea Fruit extracts could not only up-regulate the expression of OPG, but also down-regulate the expression of RANKL, and the ratio of OPG to RANKL mRNA expression in rat osteoblasts was up-regulated by Malaytea Scurfpea Fruit extracts(1.0 and 10mg/mL) significantly. In conclusion Malaytea Scurfpea Fruit ex- tracts could promote the bone formation while inhibiting bone resorption through regulating the OPG and RANKL mRNA expression of osteoblasts .
