摘要
利用藻类叶绿素a具有强荧光之特性,在激发波长418nm和发射波长680nm的条件下,发现叶绿素a浓度在0.28~88μg/L范围内与其荧光强度之间具有良好的线性关系,进而建立了直接荧光分光光度法测定水体中叶绿素a的新方法。同时以绿藻为例,建立了水样中绿藻所含叶绿素a荧光强度与绿藻生物量之间的相关关系。据此,可采用荧光分光光度法快速实时测定水样中藻类生物量,为藻类暴发的预警工作提供科学依据。将该法与传统萃取-分光光度法进行对比,无显著性差异,用于实际水样测定,结果满意。
According to the intrinsic fluorescence of chlorophyll-a,a fluorophotometric determination of chlorophyll-a in lake water was developed for the on-line monitoring of the growth of algea.The fluorophotometric measurement was carried out with excitation at 418 nm and emission at 680 nm.The fluorescnece intensity responded with the concentration of chlorophyll-a linearly in the range of 0.28 ~88 μg/L with a correlative coefficient of 0.999.By using green algae as an example,the relationship between concentration of chlorophyll-a and fluorescence intensity of green algae in water was established.Thus,the algea biovolume can be determinated rapidly by fluorophotometric to provide the scientific basis for the early warning of water bloom.Comparison was made between this method and the traditional extraction way,and no significant difference was found.The proposed method was successfully applied to the determination of real water.
