中药正肝化瘀方对肝纤维化大鼠肝组织胶原代谢的影响

Effects of Zhenggan Huayu Recipe on Collagen Metabolism of Hepatic Tissues of Hepatic Fibrosis Rats

目的:观察正肝化瘀方对肝纤维化大鼠肝组织胶原代谢的影响。方法:采用四氯化碳+高脂低蛋白饮食制备大鼠肝纤维化模型,治疗组以正肝化瘀方灌胃,正常组、模型组予以等量生理盐水灌胃,6 w后取材检测。用Jamall氏法测定各组大鼠肝组织羟脯氨酸(Hyp)含量,Western blot法检测各组大鼠肝组织α-SMA、Ⅰ型、Ⅳ型胶原蛋白的表达。结果:与模型组比较,治疗组大鼠肝组织Hyp含量及α-SMA、Ⅰ/Ⅳ型胶原蛋白表达均降低(P<0.05,P<0.01)。结论:正肝化瘀方的抗肝纤维化机制与减少肝组织胶原合成与沉积有关。

Objective：To observe effects of Zhenggan Huayu Recipe（ZGHYR）on collagen metabolism of hepatic tissues of hepatic fibrosis rats.Methods：Hepatic fibrosis rat models were built by phenixin combined with food of high fat and low protein.All rats were divided randomly into 3 groups：normal group,model group and treatment group.Rats in treatment group were given ZGHYR decoction by intragastrically administration for six weeks,while rats in normal group and model group were administered same volume normal saline.After treatment,hepatic tissue of rats was taken out,then Hyp content of hepatic tissue was detected by Jamall assay and expressions of α-SMA,collagen proteinⅠand Ⅳ of hepatic tissue were detected by Western blot assay.Results：Compared with model group,Hyp content and expressions of α-SMA,collagen proteinⅠand Ⅳ of hepatic tissue of rats in treatment group were decreased significantly（P0.05,P0.01）.Conclusions：ZGHYR decoction possesses anti-hepatic fibrosis effect through reducing production and deposition of collagen in liver tissue.
