PPP和根面脱矿对牙槽骨成骨细胞在病变牙根面附着及增殖的影响

摘要 目的观察贫血小板血浆(PPP)和根面脱矿单独及联合应用对人牙槽骨成骨细胞在病变牙根面的附着及增殖的影响。方法体外培养人牙槽骨成骨细胞,应用梯度密度离心法从人新鲜全血中获取PPP,将因重度牙周病拔除的牙齿制成病变牙根片。经PPP和EDTA脱矿分别和联合处理的病变根片作为实验组,未处理的病变根片作为对照组,分别采用细胞计数法、四唑盐比色法分析细胞在根片上的附着和增殖,扫描电镜观察PPP对根面的附着情况及其上附着的细胞形态。结果①PPP能较牢固地附着于病变根面。与对照组比较,实验组牙槽骨成骨细胞的附着和增殖显著强于对照组,(细胞附着:PPP和EDTA单独处理P<0.05,PPP和EDTA联合处理P<0.01;细胞增殖:PPP和EDTA单独处理P>0.05,PPP和EDTA联合处理P<0.05;②扫描电镜显示实验组根片上细胞显示不同程度胞体增大,胞浆突扩展,有的伸出丝状伪足。结论 PPP能牢固地附着于病变根面,PPP和根面脱矿单独和联合处理病变根片能明显加强三种细胞的附着,而PPP和根面脱矿联合处理还有显著的促细胞增殖效应。 Objective To determine whether it is possible to enhance human alveolar bone cells attachment and proliferation to diseased root surfaces of human diseased root surface treated with EDTA and freshly prepared platelet - poor plasma (PPP) from human whole blood. Methods Human alveolar ostblast bone cells (OB) grew from tissue explants and cells between the 5th and 8th passage in culture were used. PPP were obtained from fresh human whole blood by gradient density centrifugation. Periodontal - affected teeth were collected and sectioned to prepare the 4 × 4 × 1 mm 3 diseased root slices. There were four groups involving in the experiment: the untreated root slices, the root slices treated with EDTA (24%, pH = 6.7 ), the root slices treated with PPP alone and treated with EDTA and PPP. The cell attachment was assessed by cell counting and the proliferation was assessed using MTr assay. Cell morphology and growth pattern were observed by scanning electron microscopy. Results PPP could attach to the diseased root surface firmly. Cell attachment and proliferation were significantly increased in the experiment group in contrast to the control group . Most of the cells that were attached to the disease root surfaces were treated with PPP and EDTA exhibited robust and elongated with filopodial extensions using scanning electron microscopy. Conclusions PPP can attach diseased root surface firmly. Pretreatment of diseased human root surfaces with freshly prepared PPP and or EDTA may enhance /he attachment and proliferation of human alveolar bone cells. This treatment may used as a strategy for treating periodontal defects.
