摘要
利用海藻酸钠对Ca2+非常敏感的特性,成功制备了海藻酸钙负载胃蛋白酶微球。运用扫描电镜和光学显微镜对微球的形貌结构进行了分析,并对负载胃蛋白酶微球(试验组)和普通胃蛋白酶(对照组)的储存稳定性进行了测定。结果表明:随储存时间的延长,试验组的酶活力衰减幅度明显小于对照组。将试验组和对照组胃蛋白酶分别用于猪皮胶原的提取,并通过傅立叶变换红外光谱、差示扫描量热仪、凝胶电泳、激光粒度仪等检测手段,对试验组和对照组所得提取物与SigmaI型胶原进行了对比分析。结果表明:试验组和对照组所得提取物在分子质量及其分布、成分及分子结构上都与SigmaI型胶原非常相似,可确认其主要成分为胶原,且试验组的提取效率较对照组有所提高。
The calcium alginate microspheres loaded pepsin were prepared according to the property of sodium alginate’s sensitivity to Ca2+. Then, the morphological structure of the microspheres was determined. The results show that the microspheres has some nice features, and the diameter is around 1μm. Besides, the storage stability of the loaded pepsin is much better than that of blank. On these basis, the loaded pepsin were used in collagen extraction comparing to the general pepsin, and the extraction was further analyzed by fourier transform infrared spectroscopy, differential scanning calorimeter, gel electrophoresis, and laser particle size analyzer with a control of Sigma type I collagen. The results suggest that the collagen extracted by the two methods has almost the same molecular weight and distribution, structure and composition, which is similar to Sigma type I collagen. And both maintained the complete three helical structure of native collagen. Hence, the extraction could be judged as collagen on the whole. Furthermore, the results indicate that the loaded pepsin could slightly increase the productivity of collagen comparing to the general pepsin.
