摘要 目的探讨转录调控因子Twist蛋白、血管内皮生长因子(VEGF)、上皮型钙黏附蛋白(E-cad)在乳腺癌中的表达及其相关性。方法采用SP免疫组织化学法检测70例乳腺癌组织(35例癌旁组织作为对照)中Twist、VEGF、E-cad的表达,并对其临床病理特点进行分析。结果 Twist、VEGF在乳腺癌组织中的表达阳性率(72.9%、75.8%)高于癌旁组织(6.0%、8.5%),差异有统计学意义(P<0.01),E-cad在乳腺癌组织中的表达阳性率(15.7%)低于癌旁组织(71.4%),差异有统计学意义(P<0.05);Twist蛋白、VEGF阳性表达率与组织病理学分级、TNM分期、腋窝淋巴结转移有相关性(P<0.05),与患者的年龄、肿瘤直径无相关性(P<0.05);E-cad阴性表达率与患者的组织病理分级、TNM分期、腋窝淋巴结转移有相关性(P<0.05),与患者的年龄、首次肿瘤大小无相关性(P<0.05);Twist与VEGF阳性表达呈正相关,与E-cad表达呈负相关。结论 Twist和VEGF在乳腺癌组织中呈高表达,而E-cad呈低表达。Twist可能通过上调VEGF的表达,同时下调E-cad的表达,从而促进乳腺癌的浸润、转移,并在其中起重要作用。 Objective To discuss the expression of Twist,vascular endothelial growth factor(VEGF),epithelial cadherin(E-cadherin) in breast cancer and their relationship.Methods By SP immunohistochemistry we detected the Twist,VEGF,E-cad expression in 70 cases of breast cancer(35 cases of adjacent tissues as control),and with the clinical and pathological characteristics were analyzed.Results The positive rate of Twist,VEGF expression in breast cancer(72.9%,75.8%) higher than in adjacent tissues(6.0%,8.5%),the difference was statistically significant(P0.05).The positive rate of E-cad expression in breast cancer(15.7%) lower than in adjacent tissues(71.4%),the difference was statistically significant(P0.05);Twist protein,VEGF positive rate with histopathologic grade,TNM stage,axillary lymph node metastasis correlated(P0.05),with age,tumor size had no correlation(P0.05);Expression of E-cad-negative patients with pathological grade and,TNM stage,axillary lymph node metastasis correlated(P0.05),with age,the first time,no correlation between tumor size(P0.05);Twist and VEGF expression was positively correlated,Twist and E-cad expression was negatively correlated.Conclusion Twist and VEGF in breast cancer tissues showed higher expression,while lower expression of E-cad.Twist expression up regulate VEGF,while down the expression of E-cad,thus promoting breast cancer invasion,metastasis,and which may play an important role.
