摘要
目的探讨孕妇患急性早幼粒细胞白血病时全反式维甲酸(RA)治疗的安全剂量及时间点.方法 SD大鼠妊娠第8、11、14天胃饲不同浓度(20~140 mg/kg)RA,至18 d取出胚鼠观察,统计正常胚胎、畸胎、死胎或吸收胎数.结果第8天使用小量RA时胚胎正常率仅为15.79%(20 mg/kg);第11天使用大量RA时畸死胎率分别为100%(140 mg/kg)和95.12%(110 mg/kg);中小量时畸死胎率降至52.63%(50 mg/kg)和63.16%(20 mg/kg);第14天使用RA各剂量致畸、致死率大幅减小,最大量时仅为38.46%(140 mg/kg).结论 RA对神经管闭合由早期至中期、由大剂量至小剂量影响逐渐减弱,可为临床用药提供依据.
Objective To investigate the safe dose and time point of RA in treatment of pregnant women with APL.Methods Pragnant SD rats were orally given different concentration RA(20~140 mg/kg)at the 8th,11th and 14th day after pregnancy.The embryos were brought out from the pragnant SD rats at the 18th day after pregnancy,and the normal,abnormal,dead and absorptive embryos were observed and statistically analyzed.Results On the 8th day,only the pregnant rats given low dose of RA(20 mg/kg)had a few normal embryos(15.79%).On the 11th day,high dose of RA caused a lot of dead and absorptive embryos,the rate was 100%(140 mg/kg)and 95.12%(110 mg/kg),when the dose decreased,the rate was declined to 52.63%(50 mg/kg)and 63.16%(20 mg/kg).On the 14th day,all doses of RA caused a few abnormal,dead and absorptive embryos,the rate in the highest dose(140 mg/kg)was 38.46%.Conclusions The effect of RA on the neural tube closure is weakened gradually from early to late stage and from high dose to low dose.This result can provide reference for clinical medication.
