水通道蛋白4在肺腺癌细胞中的表达及其对肺腺癌细胞增殖与迁移的影响 被引量:2

摘要 目的研究水通道蛋白4(AQP4)在肺腺癌中的表达及其对肺腺癌细胞增殖与迁移的影响。方法收集20例经病理切片确诊的肺腺癌组织和4例癌周正常肺组织(距癌缘4~6cm,病理证实无肿瘤细胞),通过LSAB免疫组化法检测其中AQP4的表达。以APQ4-shRNA转染培养的肺腺癌细胞,通过Western blot法检测细胞中AQP4及E-cad-herin的表达;通过MTT法和Transwell小室法分别检测AQP4表达对肺腺癌细胞增殖与迁移的影响。结果 AQP4在肺腺癌组织中表达阳性率为55%(11/20),正常肺组织均为阴性。在肺腺癌细胞株中,AQP4均呈阳性表达且与E-cad-herin蛋白的表达呈负相关;AQP4的表达对肺腺癌细胞增殖无明显影响(P>0.05)。细胞迁移实验48h后,与对照组比较,AQP4-shRNA转染组细胞迁移显著减少(8.9%vs 14.8%,P<0.05)。结论 AQP4与肺腺癌的发生和迁移均相关;AQP4可能通过调节E-cadherin蛋白的表达而促进肺腺癌细胞的迁移。 Objective To study the correction between aquaporin 4(AQP4)and migration ability and growth of lung adenocarcinoma cells.Methods LSAB was used to examine the expression of AQP4 in 20 cases of lung adenocarcinoma tissues diagnosed pathologically and 4 cases of para-cancerous normal lung tissues.The Western blot method was used to assay expression of AQP4 and E-cadherin in lung adenocarcinoma cells which transfected with AQP4-shRNA.MTT assay was used to investigate the proliferation of the AQP4-transfected cells,and the cell migration was evaluated by using a Transwell migration assay.Results The positive expression rate of AQP4 in adenocarcinoma tissue and normal lung tissue was 55%(11 of 20)and 0% respectively.Forty-eight h after Transwell migration assay of AQP4,the migration rate in AQP4-shRNA group was reduced significantly as compared with control group(14.8% vs 8.9%,P0.05).The expression of AQP4 was positive in lung adenocarcinoma cells,and was negatively correlated with the expression of E-cadherin protein.The expression of AQP4 had no significant effect on growth and proliferation of adenocarcinoma cells(P0.05).Conclusion AQP4 is related with pathogenesis and migration of lung adenocarcinoma probably by regulating the expression of E-cadherin protein.
