摘要 目的建立快速、准确的测定南五味子有效成分五味子醇甲和五味子酯甲含量的方法.方法五味子醇甲和五味子酯甲的含量测定采用高效液相色谱法(HPLC),色谱条件:Hypersil BDSC8柱(4.6mm×150mm,5μm),检测波长254nm,柱温30℃,流速1.0mL.min-1,流动相:甲醇-0.1%醋酸(65∶35),进样量20μL.结果 HPLC测定五味子醇甲在4.950~15.43ng.μL-1(r=0.9996)范围内线性关系良好,五味子酯甲在11.38~174.6ng.μL-1(r=0.9999)范围内线性关系良好;南五味子中五味子醇甲和五味子酯甲的平均回收率分别为98.79%、99.23%,回收率的RSD分别为1.82%、2.20%(n=5).结论该方法精密度、重复性和分离效果均良好,分析快速准确,可作为南五味子中五味子醇甲和五味子酯甲的质控方法. Objective To investigate a rapid and sensitive method for determination of schisandrin and schisantherin A in Schisandra sphenanthera Rehd. et Wils. Methods To analyse Schisandrin and Schisantherin A in Schisandra sphenanthera Rehd. et Wils. by high performance liquid chromatography (HPLC) on a Hypersil BDSC8 column (4. 6 min×150 mm, 5 μm), detection wavelength was set at 254 nm and the column temperature was 30 ℃, The mobile phase was methanoi-0.1% acetic acid (65: 35), with the flow rate of 1.0mL · min-1. Results Schisandrin showed a good linear relationship at range of 4. 950-15.43ng ·μL-1 (r=0. 9996), and Schisantherin A showed a good linear relationship at range of 11.38;174. 6 ng ·μL-1 (r= 0. 9999). The average recovery rate schisandrin and schisantherin A in Schisandra sphenanthera Rehd. et Wils. were 98. 79% and 99.23;, and RSD of the method were 1.82% and 2.20; respectively; Conclusion The method is proved to be accurate, rapid, and sensitive. It can be used for quality control of schisandrin and schisantherin A in Schisandra sphenanthera Rehd. et Wils.
