摘要
目的观察喜炎平注射液对博莱霉素(bleomycin,BLM)所致小鼠肺纤维化的作用并探讨可能的机制。方法将昆明小鼠随机分为对照组、喜炎平(XYP)组、博来霉素(BLM)模型组、BLM+XYP组。经气管内1次性灌注BLM复制肺纤维化动物模型,治疗组造模后同时予尾静脉注射喜炎平直至第28天实验结束,药物干预后第7天、28天分别处死动物,第7天测定肺湿干重比(W/D),髓过氧化物酶(MPO)活性,苏木精-伊红(HE)染色观察肺泡炎症,第28天测定羟脯氨酸(HYP)含量及前Ⅰ型胶原蛋白mRNA(procollagenI mRNA)表达水平,Masson染色观察肺纤维化程度。结果第7天,BLM+XYP组小鼠较BLM组的肺泡炎程度明显减轻,肺W/D及MPO活性降低(P<0.05),第28天,BLM+XYP组小鼠肺组织HPY含量、procollagen I mRNA表达水平较BLM组明显降低(P<0.05),Masson染色显示胶原沉积减少,肺纤维化程度减轻。结论喜炎平对博莱霉素所致小鼠肺纤维化具有一定的干预作用,其机制可能与其抗炎作用有关。
Objective To observe the therapeutic effects of Xiyanping injection on bleomycin - induced pulmonary fibrosis in mice and research on its mechanism. Methods The mice were randomly divided into 4 groups: Control group, XYP group, BLM model group and BLM + XYPgroup. The BLM + XYP group were administrated bleomycin intratracheally and received an caudal vein injection of Xiyanping simultaneously, the Xiyanping injection give once a day until the end of experiment, the mice were sacrificed at the 7 th and 28 th day. The degree of lung inflammation were assessed by lung Wet / Dry ratio, myeloperoxidase (MPO) activity and Hematoxylin - eosin(HE) staining at the 7 th, the degree of pulmonary fibrosis were observed by droxyproline(HYP) contents, procollagenI mRNA expression and Masson dyeing at the 28 th. Results The BLM + XYP group show the lower lung Wet/Dry ratio and MPO activity as well as lighter aveolar inflammation compared with BLM group at the 7 th ( P 〈 0.05). The less collagen deposition , the fewer HYP contents and procollagenI mRNA expression in BLM + XYP group compared with BLM group at the 28 th( P 〈 0.05). Conclusion The Xiyanping injection could alleviate the bleomycin- induced pulmonary fibrosis in mice, The probable mechanism might be associated with the action of anti - inflammatory .
