摘要
目的:研究CD44和p-AKT蛋白在宫颈癌组织中的表达及其与临床病理学的关系。方法:应用免疫组织化学S-P法检测130例宫颈石蜡包埋组织、Western blotting法检测30例宫颈癌及癌旁宫颈组织中CD44和p-AKT蛋白的表达水平。用χ2和t检验分析CD44和p-AKT蛋白在宫颈组织中表达的意义及其与临床病理特征间的关系。结果:免疫组化结果显示在宫颈癌癌变过程中CD44和p-AKT蛋白表达阳性率呈递增趋势(P=0.001,P=0.003);CINⅢ及原位癌组与宫颈癌组CD44和p-AKT蛋白表达阳性率比较差异有统计学意义(P=0.036,P=0.048)。CD44和p-AKT蛋白表达阳性率与宫颈癌患者的年龄无明显相关性,但均与临床分期及组织学分级相关(P(CD44)=0.037,P(CD44)=0.001;P(p-AKT)=0.018,P(p-AKT)=0.002)。p-AKT蛋白的表达与盆腔淋巴结转移有关(P=0.023)。Western Blotting结果显示宫颈癌组织中CD44和p-AKT蛋白表达水平明显高于癌旁组织(P=0.002,P=0.003)。结论:CD44和p-AKT蛋白表达水平在一定程度上可能提示了从CIN向宫颈癌转化的潜能。
Objective: To research the expressions of CD44 and p - Akt protein in cervical cancer tissue and their relationships with clinicopathology. Methods: Immunohistocbemical S - P method was used to detect the expression levels of CD44 and p - Akt protein in 130 paraffin -embedded cervical specimens, Western blotting was used to detect the expression levels of CD44 and p - Akt protein in 30 specimens of cervical cancer and adjacent tissues. Chi - square test and t test were used to analyze the significances of CD44 and p - Akt protein expressions in cervical tissue and their relationships with clinicopathological characteristics. Results: The immunohistochemical resuits showed that the positive rates of CD44 and p - Akt protein expressions showed gradual increasing trends in carcinogenesis of cervical cancer ( P = 0. 001, P = 0. 003 ) ; there was significant difference in the positive rates of CD44 and p - Akt protein expressions between CIN Ⅲ plus carcinoma in situ group and cervical cancer group ( P = 0. 036, P = 0. 048 ) . There was no apparent correlation between the positive rates of CD44 and p - Akt protein expressions and the age of patients with cervical cancer, but the positive rates of CD44 and p - Akt protein expressions were correlated with clinical stages and histological grades (P(CD44) = 0. 037, P(CD44) = 0. 001 ; P(p-AKT) = 0. 018, P(p-AKT) = 0. 002) . The expression of p - Akt protein was related to pelvic lymph node metastasis ( P = 0. 023 ) . The results of Western blotting showed that the expression levels of CD44 and p - Akt protein in cervical cancer tissue were significantly higher than those in adjacent tissue ( P = 0.002, P = 0.003 ) . Conclusion: The levels of CD44 and p - Akt protein may indicate the potential of CIN transforming to cervical cancer to a certain extent.
