摘要
采用溶剂萃取的方法,制备了白皮锦鸡儿(Caragana leucophloea Pojark.)地上部分各提取物组分,以乙酸乙酯萃取组分的黄酮含量最高、抗氧化活性最强。乙酸乙酯萃取组分继续用高速逆流色谱制备各峰组分,其中逆流峰4组分中黄酮化合物含量为20.51%,是乙酸乙酯萃取组分中黄酮含量(3.90%)的5.26倍。逆流峰4组分对DPPH和β-胡萝卜素-亚油酸氧化的半抑制浓度(Ic卯)分别为28.3μg/mL和54.8p.g/mL。结果表明溶剂萃取并结合高速逆流色谱能有效地制备白皮锦鸡儿黄酮。
The solvent fractions were prepared from the crude ethanol extract of the aerial parts of Garagana leucophloea Pojark. (Leguminosae) by organic solvent fractionstions. Both the high flavonoid content and strong antioxidant activity were examined in the ethyl acetate fraction which was further purified by high-speed counter-current chromatography (HSCCC). Of the HSCCC peak fractions,the strongest antioxidant activity with ICso value as 28.3 μg/mL on DPPH inhibition or IC50 value as 54.8 μg/mL onβ-carotcne-linolcic acid bleaching was found in HSCCC peak 4 fraction, which flavonoid content (20.51%) was also the highest that was 5.26 fold of the ethyl acetate fraction (3.90 % ). Solvent fractionation combining with HSCCC may be a favorable method for C. leucophloea flavonoid preparation.
