摘要
建立高效液相色谱法测定黑豆皮提取物中矢车菊素-3.0.葡萄糖苷的含量。色谱条件:色谱柱为Inertsil ODS-3150×4.6mm 5um;流动相为:10%甲酸溶液(A).甲醇(B),梯度洗脱程序:0-20minB由5%-60%,20—25rainB60%^-100%,25~30minB100%;流速1.0mL/min;检测波长520nm;柱温30℃;进样量10汕。线性范围为0.01245mg/mL~0.1992mg/mL(r=1),加标平均回收率为96.86%,RSD为1.10%(n=6)。本方法准确度高、精密度高、重复性好、简捷易操作,可以作为黑豆皮提取物中矢车菊素-3-O-葡萄糖苷含量的测定方法。
This article is to establish a HPLC method fdr analyzing content of cyanidin-3-O-glucoside in blgck soybean coat extract. The chromatographic conditions are as follows:column Inertsil ODS-3 150 × 4.6 mm 5 urn;mobile phase A = water + 10% formic acid solution,B = methanol (0 -20 min linear gradient from 5% to 60% B and 20 -25 min linear gradient from 60% to 100% B then 25 - 30 min,isocratic 100% B) ;flow rate 1.0 mL ·min-1 ;detection wavelength 520 nm ;column temperature 30℃ ;injection volume 10 μL. The method is liner within the range of 0. 01245 mg/mL -0. 1992mg/mL (r= 1) ,average recovery is 96.86% ,RSD of 1.10% (n =6). This method is accurate,precise, repeated and easy to operate. It is equaled for analyzing content of cyanidin-3-O-glucoside in black soybean coat extract.
