摘要
本文研究了D-101大孔树脂纯化无患子总皂苷的工艺条件。通过考察上柱样品量、吸附时间、洗脱剂浓度、洗脱剂用量、洗脱速度等因素的影响,确定了较优的工艺方案:30g大孔树脂柱(2cm×40cm)上样20mL,吸附30min,200mL水洗除杂,300mL50%乙醇以3BV/h的速度洗脱。经过大孔吸附树脂富集、纯化后,无患子总皂苷纯度可达64%以上,说明采用本法富集、纯化无患子总皂苷可行。
To study the technological parameters of the purification process of total saponins of Sapindus mukorossi Gaertn with macroreticular resin. After adsorptive characteristics and elutive parameters of the process such as sample volume, adsorbtion time, eluant concentration, eluant volume, eluant velocity were studied, the 20 mL of extractive of total sapo- nins of S. mukorossi was purified with a column of macroreticular rosin (2 cm x 40 cm, dried weight 30 g) and was washed with 200 mL of distilled water,then was eluted with 300 mL of 50% ethanol in the eluant velocity of 3 BV/h. With macroreticular rosin to adsorb and purify,the purity of total saponins of S. mukorossi roached 64%. The process of applying macrooroticular rosin to adsorb and purify total saponins of S. mukorossi was feasible.
