摘要
运用荧光光谱、紫外吸收光谱、红外光谱等研究了槲皮素(Quercetin,Que)与BSA的结合作用,并从Eu3+与Que的竞争作用、热力学参数的变化、配位化合物的生成三个方面分析了Que与BSA结合作用的影响因素。结果表明,Que对BSA的荧光光谱具有猝灭作用,其猝灭机制为静态猝灭,BSA发射峰显著蓝移;Eu3+的存在使得BSA发射峰蓝移程度降低,猝灭常数、结合常数、结合位点数减小,但没有改变Que与BSA的作用机制;在较大浓度Eu3+存在下,Que与BSA作用的相关系数减小。
The interaction between quercetin(Que) and bovine serum albumin(BSA) in the presence of Eu3+ had been investigated by the fluorescence, ultraviolet absorption and infrared spectra. The fluorescence of BSA was quenched by Que, and the quenching mechanism was a static quenching process. The maximum emission wavelength of BSA was appreciable blue-shift. When appropriate amount of Eu3+ was added to the mixture of Que and BSA,a smaller blue-shift of the maximum emission wavelength of BSA was observed,and the quench- ing constant, binding constant,number of binding site and correlation coefficient (R) decreased. But the quench- ing mechanism of BSA by Que was not changed in the presence of Eu3+. The analysis indicated that the effect of Eu3+ on the interaction between Que and BSA was related to the Eu3+ competition role, the change of thermody- namic narameters and the Eua+ complex formation in aqueous solution.
