摘要
金属离子可以影响纤维素内、外切酶的活性,且不同金属离子对其活性影响程度也不同。据此,本实验选取三种纤维素酶进行单因素实验,对其内、外切酶活性进行研究。实验结果表明,当金属离子的质量浓度为:K+0.9 mg/mL,Ca2+0.3 mg/mL,Zn2+1.2 mg/mL,Cu2+1.2 mg/mL,能较好地提高纤维素内切酶活;而Mg2+﹑Ca2+﹑Ba2+﹑Cu2+﹑Zn2对纤维素外切酶活都有抑制作用;Al3+对纤维素内、外切酶活都有抑制作用;不同价态阴离子对纤维素内、外切酶活影响不大。
Because of the metal ions can affect the activity of endo-l,4-13-D-glucanase (EG) and exo-1,4-β-D-glucanase (CBH) on cellulose and different metal ions have the different activity. Accordingly, we select three kinds of cellulase and do the single-factor experiment. The activity of EG and CBH on cellulase were results showed: The activity of EG can improve effectively when the concentration of K+ is 0.9 mg/mL, Ca2+ 0.3 mg/mL, Zn2+ 1.2 mg/mL, Cu2+ 1.2 mg/mL; Mg2+, Ca2+, Ba2+, Cu2+, Zn2+ showed the inhibitory character. The Al3+ had some inhibition on the cellulase activity of EG and CBH. Anions with different valences have no effect on the activity of EG and CBH.
