摘要 目的探讨不同诱导方法对人外周血γδT细胞生成率的影响。方法用抗γδT细胞受体,IL-2,人肝癌细胞SMMC-7721提取的热休克蛋白70(HSP70)以及它们的不同组合诱导人外周血γδT细胞的生成。结果γδT cell re-ceptor(0.4μg/ml)即可诱导γδT细胞的大量产生,达61.5%,在0.8μg/ml时进一步升高,达75.7%;当与IL-2联合应用时产生率无明显变化;在与HSP70联合使用时,产量明显升高,分别达71.1%和85.6%;HSP70与IL-2联合使用也可以产生大量γδT细胞,达75.6%。结论用抗γδT细胞受体,IL-2,人肝癌细胞SMMC-7721提取的HSP70以及它们的不同组合可以诱导人外周血γδT细胞的大量生成。 Objective Discussing the influence on producing rate of peripheral γδ T cells with different inductive methods.Methods γδT cells receptor resistance,IL-2,HSP70 extracted from SMMC-7721 hepatumor cells and their different combination can be used to induce human peripheral γδT lymphocytes.Results γδT cells receptor(0.4 ug/ml) can induce large numbers of γδT cells,to 61.5%,and when it is 0.8 ug/ ml,there are further more γδT cells,increasing to 75.7%;Producing rate of γδT cells did not change when combined with IL-2,but the yield was significantly higher when combined with HSP70,respectively,to 71.1% and 85.6%,combined therapy that using HSP70 and IL-2 also can produce a lot of γδT cells,to75.6%.Conclusion γδT cells receptor resistance,IL-2,HSP70 extracted from SMMC-7721 hepatumor cells and their different combination can induce a large number of γδT cells from human peripheral blood.
