尾加压素Ⅱ在大鼠心肌纤维化中的表达及作用 被引量:1

摘要 目的研究尾加压素Ⅱ(urotensinⅡ,UII)在大鼠心肌纤维化(myocardial fibrosis,MF)中的表达及作用。方法注射盐酸异丙肾上腺素(isoproterenol,Iso)15 mg.kg-1复制大鼠MF模型。Masson三色染色分析大鼠心肌细胞外基质沉积情况,免疫组化染色及RT-PCR评估UII基因和蛋白表达情况。结果与正常对照组比较,注射Iso后12 h,UIImRNA及蛋白表达即明显增加(P<0.01),其中mRNA表达1周后有所回落,但3周后仍未降至正常(P<0.05),而UII蛋白表达水平仍然随着纤维化程度的加重稳步增高。结论 UII的表达在缺血性坏死心肌中明显增加,提示UII的表达与纤维化程度有关联,可能参与了MF的过程。 Objective To investigate the expression and effect of UII in rat myocardial fibrosis.Methods Rat myocardium necrosis model was established by isoproterenol(Iso) injection(15 mg.kg-1).The extracellular matrix change of myocardium was analyzed by Masson stain,the expression of UII gene and protein were analyzed by immunohistochemistry and RT-PCR.Results Compared with control group,the gene and protein expression of UII were increased.UII mRNA arrived peak at 12 h(P0.01) and began decreased at 1w,but still higher than normal level until 3 w(P0.05).However,UII protein increased steadily according with the fibrosis severity.Conclusion UII expression levels are increased significantly in myocardial ischemia and the increasing of UII expression has a closed relation with fibrosis,which indicates UII may be involved in the process of myocardial fibrosis.
