摘要
利用氯乙酸化、酰胺化等方法合成了大分子多糖阿拉伯半乳聚糖修饰的DOTA钆配合物,采用红外光谱测试、13C NMR、元素分析及ICP-AES手段进行了表征,测试其在水中及牛血清白蛋白溶液中的弛豫效率。结果表明,其弛豫效率远远高于目前临床应用的Gd-DOTA与Gd-DTPA,是比较好的潜在的磁共振成像造影剂。
Arabinogalactan derivatives conjugated with gadolinium-1,4,7,10-tetraazacyclododeca ne-N,N′,N″,N-tetraacetic acid(Gd-DOTA) by ethylenediamine(Gd-DOTA-AGCMC6) has been synthesized and characterized by means of Fourier Transform Infrared Spectra(FTIR),1H nuclear Magnetic Resonance(1H NMR),and inductively coupled plasma atomic emission spectrometry(ICP-AES).The T1 relaxivity of Gd-DOTA-AGCMC6 for 7.64 mmol^-1·L·s^-1 in D2O,7.78 mmol^-1·L·s^-1 in 0.725 mmol^-1·L BSA were higher than that of commercial MRI contrast agent(Gd-DOTA) in aqueous solution.The results indicate that the arabinogalactan-bound complexe is a potential contrast agent for MRI.
