绝经后妇女基质金属蛋白酶-3和抑制因子与骨密度和骨保护蛋白及其配体的关系

Relationship of Postmenopausal Circulating Matrix Metalloproteinase-3 and Its Inhibitor Levels with BMD and Osteoprotegrin and Its Ligand

摘要 目的:探讨绝经后妇女血清基质金属蛋白酶-3(MMP-3)和抑制因子(TIMP-1)浓度与骨密度(BMD)和骨保护蛋白(OPG)及其配体(OPGL)的关系。方法:120例绝经后妇女用双能X线吸收法(DEXA)测定腰椎正位,股骨颈,华氏区和大粗隆的BMD,根据BMD检测结果分为正常组(A组)、低骨量组(B组)和骨质疏松组(C组)各40例,用酶联免疫吸附试验(ELISA)测定血清MMP-3、TIMP-1和OPG及OPGL浓度,计算MMP-3/TIMP-1、OPG/OPGL比值;结果:①C组MMP-3浓度高于A组(153±121ng/ml与125±101ng/ml,P<0.05),而TIMP-1的数值略低于A组(134±106μg/L与147±130μg/L)。②C组中MMP-3与MMP-3/TIMP-1比值与血清OPG存在明显正相关性,MMP-3/TIMP-1比值与BMD和OPGL存在明显负相关性(均P<0.05),校正年龄和体质量指数后,以上数据相关性增高(P<0.05)。结论:绝经后骨质疏松症(PMOP)患者MMP-3/TIMP-1比值与BMD和OPG和OPGL具有关联性,MMP-3/TIMP-1比值升高可能为PMOP伴随骨代谢转换过程增快的表现。 Objective: To study the serum matrix metalloproteinase-3 (MMP-3) and tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) and their correlations with osteoprotegrin (OPG) and its ligand (OPGL) and bone mineral density (BMD) in aged postmenopausal Chinese women. Methods: The serum MMP-3, TIMP-1, OPG and OPGL in 120 postmenopausal Chinese female volunteers were measured by using ELISA. The ratio of MMP-3 to TIMP-1 was calculated. BMD was measured by using dual energy X-ray absorptiometry. Results: (1) Serum MMP- 3 concentrations were significantly higher in osteoporosis (153 ± 121 ng/mL) than in age-matched normal controls (125 ± 101 ng/mL) ; (2) In osteoporosis, there were notably negative correlations between ratio of MMP-3/TIMP- 1 and BMD and OPGL (P〈0.05) as well as positive correlations between MMP-3, ratio of MMP-3/TIMP-1 and sOPG (P〈0.05). Conclusion: There are significant correlations between ratio of MMP-3 and TI.MP-1 and sOPG and BMD, and MMP-3 may be increased with increases in bone-metabolism. The increases of ratio of MMP-3/ TIMP-1 appear possibly as a concomitant event in high bone turnover state, such as postmenopausal osteoporosis.
