摘要
目的观察结扎冠状动脉左前降支造成心肌缺血时大鼠小肠内孤啡肽(orphanin FQ)的表达变化。方法 54只雄性SD大鼠按随机数字表法分为三组:①正常对照组(n=6),麻醉后直接取材;②假手术组(sham组,n=24),只开胸暴露心脏不结扎冠脉;③结扎冠状动脉组(coronary artery occlusion group,CAO组,n=24),单纯扎闭冠状动脉左前降支。其中,sham组和CAO组又分为4个亚组:0.5 h组、1 h组、3 h组、6 h组,每亚组大鼠均为6只。采用免疫组化观察并分析结扎大鼠冠状动脉不同时点小肠组织内OFQ的表达变化情况;应用酶联免疫吸附技术对比分析不同时点小肠组织内OFQ的表达水平。结果免疫组化半定量分析结果显示,CAO后各时点(除0.5 h外)小肠黏膜和肌间神经丛OFQ的水平较正常对照组和同时点sham组显著升高(P<0.05);酶联免疫吸附技术结果显示CAO组小肠OFQ的水平较同时点sham组显著升高(P<0.05),CAO 3 h组表达达到高峰,与sham组比较差异有统计学意义(P<0.01)。结论急性心肌缺血时大鼠小肠内孤啡肽的表达增加,孤啡肽可能作为一种肠神经递质或调质来影响胃肠运动和分泌。
Objective To investigate the expression of orphanin FQ(OFQ) in intestine tissues of rats with acute myocardial ischemia.Methods Fifty-four healthy adult male SD rats weighed 260-280 g were randomly divided into three groups:normal control group,sham group and coronary artery occlusion(CAO) group.The rats in sham group and CAO group were subgrouped into 0.5 h,1 h,3 h,6 h groups.Immunohistochemical SABC staining and ELISA were used to observe the expression of OFQ in the intestine tissues after coronary artery occlusion.Results Semi-quantitative immunohistochemical analysis showed that the levels of OFQ in CAO subgroups were significantly higher than in sham subgroups and control group(P〈0.05).The results of ELISA showed that the levels of OFQ were significantly higher in CAO group than in sham group at the same time point(P〈0.05 or P〈0.01),and reached the peak at 3 h in CAO group.Conclusion Expression of OFQ significantly increases in intestine tissues of coronary artery occlusion rats.OFQ may act as a neurotransmitter in the gastrointestinal motility and secretion.
