摘要
目的建立治咽颗粒的质量控制方法。方法采用TLC法鉴别金银花、甘草。采用HPLC法测定绿原酸的含量,色谱柱为Zorba SB-Aq-ODS(150 mm×4.6 mm,5μm),流动相为乙腈∶0.025%磷酸溶液(13∶87),检测波长为327 nm,柱温为25℃,流速为1 mL/min,进样量10μL,以外标法测定治咽颗粒中绿原酸含量。结果金银花、甘草TLC鉴别色谱特征明显,专属性强;绿原酸在28.8～67.2μg/mL范围内线性关系良好,相关系数为0.999 9,平均回收率为98.6%(n=6,RSD=1.37%)。结论该方法可用于治咽颗粒的质量控制。
Objective To develop a method for the quality control of zhiyan granules.Methods The honeysuckle flower and Radix Glycyrrhizae in zhiyan granules were identified respectively by TLC.The content of chlorogenic acid in zhiyan granules was determined by HPLC.An Zorba SB-Aq-ODS was used with a mobile phase consisted of mixture of acetonitrile-0.025% phosphoric acid（13∶ 87）,the flow rate was 1.0 mL/min.The UV detective wavelength was at 327 nm.Results The TLC identification for herbs Honeysuckle Flower and Radix Glycyrrhizae was obvious and highly specific.The calibration curve was liner within the range of 28.8~67.2 μg/mL for chlorogenic acid（r=0.999 9）.The average recoveries of chlorogenic acid was 98.6%（n=6,RSD=1.37%）.Conclusion The method may be served as the quality control of zhiyan granules.
