摘要
目的:研究龙牡壮骨颗粒中黄芪甲苷的含量测定方法。方法:高效液相色谱-蒸发光散射检测法。色谱柱为Diamonsil C18,4.6mm×250mm,5μm;流动相为甲醇-水(75:25);流速为1.0mL·min-1;柱温为30℃;检测器为Varian380-LC型蒸发光散射检测器。结果:该方法能很好分离被测组分与其他杂质,在实验浓度范围内峰面积的对数值与浓度的对数值呈良好的线性关系,HPLC-ELSD法测定的线性范围为0.2~1mg·mL-1;相关系数r=0.9998;样品平均回收率为99.5%(n=9);RSD=0.23%。结论:本法简便、可靠,专属性强,重复性好,可用于测定龙牡壮骨颗粒中黄芪甲苷的含量。
Objective: To develop a method for the determination of Astragaloside IV in Longmu Zhuanggu Granules. Methods:Method Using high performance liquid chromatography (HPLC) - evaporative light scattering Detector assay for content determination. For Diamonsil C18 column and 4. 6mm × 250mm, 5 μm, Mobile phase for Methanol - water (75:25), Velocity 1.0 mL·min^-1, Column temperature for 30℃, Varian 380 - LC type evaporative light scattering detector. Results:This method can be measured with other components are separated in the experiment, the concentration of impurity peak area within the scope of the concentration of numerical value and a good linear relationship, the linear range of 0. 2 ·1mg · mL^-1, r = 0. 9998,the average recovery was 99.5 per cent ( n = 9 ), RSD = 0. 23%. Conclusion: This method is simple, reliable, strong specificity, reproducible and can be used to assays the content determination of Astragaloside IV in Longmu Zhuanggu Granules.
