中药材贝母DNA不同提取方法的比较

Comparative Study of Different Extraction Methods for the Isolation of DNA from Traditional Chinese Medicine Fritillaria

目的探讨中药材贝母DNA的不同提取方法,为中药贝母的分子生物学鉴定提供参考.方法利用苯酚法、CTAB法、SDS法从中药贝母中提取DNA,通过紫外分光光度和琼脂糖凝胶电泳对所得DNA样品含量及纯度进行检测.结果利用3种不同的方法提取,药材贝母DNA的A260/A280在1.80±0.23左右.结论利用3种不同DNA提取方法,均可从贝母样本中提取得到DNA,且CTAB法提取贝母的DNA含量高、纯度好.

Objective To discuss the different extraction methods for the isolation of DNA from traditional Chinese medicine fritillaria and provide the reference for the molecular biology appraisal of the traditional Chinese medicine fritillaria.Method The Phenol,CTAB and SDS methods were used to isolate DNA from the traditional Chinese medicine fritillaria,and ultraviolet spectrophotometry and agarose gel electrophoresis were used to examine DNA content and quality.Results The ratios of A260/A280 for DNA were between 1.80±0.23 with three different extraction methods.Conclusion The three methods can be used for the extraction of DNA from fritillaria.The CTAB method is the best method for the extraction of DNA from fritillaria,and the DNA with highest content and purity from fritillaria can be got with this method.
