摘要
目的探讨雌激素联合甘草酸对去卵巢小鼠骨代谢及生物力学的影响。方法将60只小鼠随机分为6组,假手术组不作任何处理,余5组行去卵巢手术,术后4 d无异常开始灌胃给药。去卵巢组不予任何药物,雌激素组予32μg/(kg.d)雌激素,低剂量甘草酸组予50μg/(kg.d)甘草酸,高剂量甘草酸组予100μg/(kg.d)甘草酸,联合组予32μg/(kg.d)雌激素和50μg/(kg.d)甘草酸,给药45 d后处死小鼠。采用分析天平称量小鼠器官并计算内脏指数,检测血清钙及碱性磷酸酶(ALP)水平,化学比色法检测骨组织羟脯氨酸(Hyp)水平,计算股骨断裂载荷、最大载荷及第四腰椎最大载荷。结果低剂量甘草酸组子宫指数明显低于假手术组及雌激素组,Hyp高于去卵巢组;高剂量甘草酸组第四腰椎最大载荷明显低于雌激素组;联合组子宫指数、第四腰椎最大载荷明显高于去卵巢组,第四腰椎最大载荷明显高于假手术组;去卵巢组子宫指数低于假手术组,子宫指数、胸腺指数低于雌激素组;雌激素组子宫指数、第四腰椎最大载荷高于去卵巢组,胸腺指数低于去卵巢组,第四腰椎最大载荷高于假手术组;P<0.05或0.01。结论甘草酸单用或联用雌激素对骨质疏松有一定的防治作用。
Objective To investigate the effects of combination of estrogen and glycyrrhizic acid(GA) on bone metabolism and biomechanic in ovariectomized mice.Methods Sixty female mice were randomly divided into six groups,all mice were subjected to bilateral ovariectomy except sham operation group,then mice showed no abnormal behavior were administrated intragastrically with drugs 4 d after.The ovariectomized group was given vehicle,the estrogen group was given estrogen of 32 μg/(kg·d),the low dose of GA(LGA) group was given GA of 50 μg/(kg·d),the high dose of GA(HGA) group was given GA of 100 μg/(kg·d),the combination(ELGA)group was given estrogen of 32 μg/(kg·d)and GA of 50 μg/(kg·d).After 45 days,the viscera index,the content of serum Ca2+,ALP and bone hydroxyproline(Hyp),biomechanical properties of bone(femur and lumbar)were measured.Results The ovary index in LGA group was significantly lower than that in sham-operation group and estrogen group,and the bone Hyp was higher than that in ovariectomized group;the maximum load of the fourth vertebra in the HGA group was significantly lower than that in estrogen group;the ovary index and the maximum load of the fourth vertebra in the ELGA group were significantly higher than those in ovariectomized group,and the maximum load of the fourth vertebra was significantly higher than that in sham-operation group;the ovary index in ovariectomized group was lower than that in sham-operation group,the ovary index and thymus index were lower than those in estrogen group;the ovary index and the maximum load of the fourth vertebra in estrogen group were higher than those in ovariectomized group,the later index was higher than that in sham-operation group,P0.05 or 0.01.Conclusions Both GA single use and its estrogen combination therapy have preventive effect in osteoporosis.
