一株嗜热菌的分离、鉴定及海藻糖合成酶基因的克隆

摘要 从广西贺州市80℃热泉分离了一株嗜热菌TTH,生长温度范围在55℃~85℃,最适生长温度70℃左右,最高生长温度85℃,G+C物质的量分数为67.6%.对菌株TTH从形态、培养条件、16S rRNA基因序列和系统发育分析方面进行了研究,表明该菌株与3株嗜热栖热菌(Thermus thermophilus)的同源性达到98%,初步鉴定是嗜热栖热菌(Thermus thermophilus).克隆了该菌的海藻糖合成酶基因,它与菌株Thermus thermophilus RQ-1,Thermus caldophilus,Thermus thermophilus HB8,Thermus thermophilus的同源性达到87%. A thermophile strain, designated as TI'H, was isolated from 80℃ hot spring at Hezhou county in Guangxi province. It could grow at temperatures from 55℃ to 85℃, with the optimal temperature 70℃, the highest temperature 85℃, and with 67. 6% molar content of the G + C. The researches which were based on the TFH morphological, culture conditions, 16S rDNA sequence, as well as the phylogenetic analysis show that the strain TTH has 98% sequence identity with three kinds of Thermus thermophilus. Therefore the strain is tentatively identified as Thermus thermophilus. Trehalose synthase gene of TTH was cloned and analyzed. It has 87% identity to the treS gene from Thermus thermophilus RQ-1, Thermus caldophilus, Thermus thermophilus HB8 and Thermus thermophilus.
