姜黄素与银杏内酯B联合三氧化二砷在治疗白血病中对心肌细胞氧化损伤保护的作用比较 被引量:2

摘要 目的研究比较银杏内酯B与姜黄素联合三氧化二砷(As2O3)治疗白血病过程中对心肌细胞氧化损伤的保护作用。方法对人白血病细胞株NB4进行培养,设立对照组、As2O3组、银杏内酯B组及姜黄素组。对照组:原营养液继续培养;As2O3组:加入As2O32μmol/L;银杏内酯B组:加入As2O32μmol/L+银杏内酯B 0.1mmol/L;姜黄素组:加入As2O30.5μmol/L+姜黄素50μmol/L。以分光光度法检测超氧化物岐化酶(SOD)、谷胱甘肽过氧化酶(GSH-PX),流式细胞仪检测细胞内活性氧(ROS)水平。结果经姜黄素、银杏内酯B干预后的NB4细胞超微结构都能得到明显改善,姜黄素、银杏内酯B均显著提高了NB4细胞内ROS、SOD含量及GSH-PX活力,姜黄素组较银杏内酯B组显著提高NB4细胞内ROS、SOD含量及GSH-PX活力(P<0.01)。结论姜黄素、银杏内酯B均可联合As2O3,上调机体ROS、SOD水平,提高GSH-PX的活性,拮抗自身氧化损伤,姜黄素对心肌细胞氧化损伤的保护作用优于银杏内酯B,为进一步提高白血病临床疗效打开了新思路。 Objective To compare the effect to oxidative damage of myocardial cell by the combination of ginkgolide B with As2O3 and the combination of curcumin with As2O3 for the treatment of leukemia. Methods The NB4 cells were cultured and divided into control group,As2O3 group,ginkgolide B group and curcumin group.Control group was continuously cultured with the original medium,As2O3 group was added with As2O3 2 μmol/L,ginkgolide B group was added with As2O3 2 μmol/L + ginkgolide B 0.1 mmol/L,curcumin group was added with As2O3 0.5 μmol/L+curcumin 50 μmol/L.The amounts of superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) were tested by spectrophotometry.The expression of reactive oxygen species(ROS) was tested by flow cytometry. Results The intervention by curcumin or ginkgolide B improved significantly the ultrastructure of NB cells.Curcumin or ginkgolide B either raised significantly ROS,SOD contents and GSH-PX activity,and the efficacy was higher by the combination of curcumin with As2O3 than by the combination of ginkgolide B with As2O3. Conclusion The combination of curcumin with As2O3 can antagonize oxidative damage of organism and provide a new idea of clinical efficacy of leukemia.
