摘要
目的探讨六味地黄丸对盐酸二甲双胍(MET)在大鼠体内药代动力学的影响。方法将10只大鼠随机分为联合用药组和MET组,联合用药组灌胃六味地黄丸7d后再给予MET;MET组大鼠直接给予MET,于给药后不同时间采集血样,高效液相色谱法测定MET的血药浓度,绘制药-时曲线,用DAS 2.0软件计算药代动力学参数。结果 MET组、联合用药组的药代动力学参数差异有统计学意义(P<0.05),半衰期分别为(3.25±0.51)、(5.03±0.67)h;0~10 h的血药浓度-时间曲线下面积分别为(129.52±5.99)、(142.81±5.79)mg/(L·h);药物全部消除时的血药浓度-时间曲线下面积分别为(159.49±12.85)、(200.39±16.92)mg/(L·h);达峰时间分别为(1.67±0.29)、(3.0±0.82)h。结论联合用药后六味地黄丸对MET的药代动力学过程有显著影响。
Objective To study the effects of co-administration Liuwei dihuang pills on the pharmacokinetics of metformin hydrochloride in rats. Methods 10 rats were randomly divided into 2 equal groups. After the rats of the co-administration group were treated with Liuwei dihuang pills for 7 days, both groups were orally administrated metformin hydrochloride. The blood samples were collected at different time, and the concentrations of metformin hydrochloride in plasma were determined by high performance liquid chromatography. According to the curve of concentration-time, the software of DAS 2.0 was used to calculate the pharmacokinetic parameters. Results The significantly changed pharmacokinetic parameters of the controlling group and the testing group (P 〈0.05) had statistical significance: half life were (3.25±0.51), (5.03±0.67) h, respectively; area under the plasma concentration-time curve from 0 to 10 h were (129.52±5.99), (142.81±5.79) mg/(L.h); area under the plasma concentration-time curve from 0 h extrapolated to infinity were (159.49±12.85), (200.39±16.92) mg/(L.h); peak time were (1.67±.29), (3.0±0.82) h, respectively. Conclusion Liuwei dihuang pills has significant effect on the pharmacokinetic process of metformin hydrochloride in rats.
