摘要
目的:研究骨化三醇对佐剂性关节炎大鼠模型血清中TNF-a、IL-4和IL-10表达水平的影响,探讨骨化三醇对佐剂性关节炎的治疗作用及相关机制。方法:雌性大鼠45只,随机分为正常对照组,佐剂性关节炎模型组,骨化三醇治疗组,每组15只,2周后,处死全部大鼠,断头取血,检测血清中TNF-a、IL-4和IL-10表达水平。结果:模型组与正常组比较,血清TNF-a水平升高,IL-4、IL-10水平降低,治疗组与模型组比较血清TNF-a表达水平明显降低,IL-4、IL-10表达水平上调。结论:骨化三醇降低佐剂性关节炎大鼠血清TNF-a的表达,上调IL-4、IL-10的表达水平,对类风湿性关节炎的治疗有一定意义。
Objective: To study the effects of 1,25--droxyvitamin D3 on serum TNF--a,IL-4 and IL- 10 in adjuvant arthritis rats and explore its treatment function and related mechanisms. Methods: All 45 rats were randomized into 3 groups: normal control group, adjuvant arthritis group and 1,25--droxyvitamin D3 therapy adjuvant arthritis group, with 15 in each. The expression levels of TNF-a, IL-4 and IL-10 in serum after 2-- week treatment were detected. Results:TNF-a level was higher and IL-4 and IL-10 levels were lower in adjuvant arthritis group than those in normal control group (P 〈(0.05). The expression level of TNF-a in the 1,25-droxyvitamin D3-treated group was significantly lower than those in 1,25--droxyvita- rain D3 therapy adjuvant arthritis group, but the expression levels of IL- 4 and IL- 10 were increased (P〈0.05). ConclusiOn: 1,25-droxyvitamin D3 therapy down--regulates the expression level of TNF--a and up--regulates the levels of IL-4 and IL-10, showing certain significance in the treatment of rheumatoid arthritis.
