豚鼠COPD模型MMP-9和TIMP-1的表达及吸入糖皮质激素对其表达的影响

摘要 目的探讨豚鼠COPD模型MMP-9和TIMP-1的表达,以及布地奈德干预后MMP-9与TIMP-1表达的变化。方法健康豚鼠30只随机分为3组:正常对照组、模型组、布地奈德组。用香烟熏吸建立豚鼠COPD模型,HE切片观察肺组织病理学改变,肺组织炎症程度的变化,应用免疫组化检测肺组织中MMP-9与TIMP-1的表达,以及药物对它们的影响。结果 COPD模型组豚鼠肺组织内MMP-9、TIMP-1表达均高于正常对照组(P<0.01);布地奈德组MMP-9表达比模型组明显下降,但仍较正常对照组高(P<0.05),而布地奈德组与模型组之间TIMP-1表达差异无统计学意义(P>0.05)。结论 MMP-9表达增强,使细胞外降解增加,可能是COPD形成机制之一。糖皮质激素可降低豚鼠COPD模型MMP-9的表达,可能延缓COPD的形成,但对TIMP-1表达无影响。
