磺胺母核人工抗原的制备

摘要 吡啶为溶剂,用对氨基苯环酰胺和丁二酸酐为原料合成磺胺母核的半抗原(H),产物结构通过HNMR确证。再用牛血清白蛋白(BSA)通过碳二亚胺法合成具有诱导产生抗体能力的免疫原(H-BSA),通过紫外光谱定性测定以及间接ELISA检测H-BSA证明成功与否。磺胺母核的半抗原合成的最佳实验条件是:温度60℃,溶剂选用吡啶,反应时间6 h,加热方式为油浴加热。以期用H-BSA免疫家兔,制备多克隆抗体,为胶体金研究奠定基础。 The hapten of sulfonamides(H) was synthesized by using sulfanilamide and succinic anhydride where pyridine is used as dissolvent,which is proved by HNMR.Moreover,H-BSA was synthesized by catodiimide method with Bovine Serum Albumin(BSA) and H.And the H-BSA can be proved by UV scan and indirect ELISA method that whether it was coupling success.The optimal experiment condition for preparation of H is using pyridine as dissolvent and heated with oil bath at temperature 60 ℃ for six hours.With the hope that we could get polyclonal antibody by immunizing rabbits with H-BSA.It will lay the foundations for the research of colloidal gold.
