掌叶半夏提取物的有效部位对宫颈癌细胞株HPVE6和P53基因表达的影响 被引量:12

Effects of Pinellia extract fraction on expressions of HPVE6 and P53 genes in cervical cancer cell lines

摘要 目的:观察中药掌叶半夏提取物的有效部位(PE)对CaSki和HeLa细胞人乳头瘤病毒(HPV)E6基因和P53基因表达的影响,进一步阐明PE的抑癌机制。方法:0.15mg/mL的PE作用于CaSki和HeLa细胞24h后,采用RT-PCR方法检测两种细胞HPVE6和P53 mRNA表达的变化,采用Western Blot检测CaSki和HeLa细胞HPVE6、P53蛋白表达的变化。结果:0.15mg/mL的PE作用24h后CaSki和HeLa细胞HPVE6 mRNA和蛋白的表达均明显下降,而P53mRNA和蛋白的表达均明显上升(P<0.05,P<0.01)。结论:PE可以明显抑制CaSki和HeLa宫颈癌细胞株HPVE6基因的表达、促进其P53基因的表达,这可能是PE抗宫颈癌作用的关键环节。 Objective: To investigate the mechanisms of inhibition effect of Pinellia extract fraction(PE) on CaSki and HeLa cervical cancer cell lines cultured in vitro,observe the effects of PE on expressions of human papilloma viruse(HPV)E6 and P53 in these two kinds of cells.Methods: Expressions of HPVE6 and P53 mRNA were detected by RT-PCR,and expressions of HPVE6 and P53 proteins were detected by western blot assay.Results: Expressions of HPVE6 mRNA and protein were down-regulated,while expressions of P53 mRNA and protein were up-regulated significantly after treatment with 0.15mg/mL PE for 24 hours.Conclusion: PE can inhibit growth of CaSki and HeLa cells by down-regulating expression of HPVE6 and up-regulating expression of P53.This may be the key point of the anti-cervical cancer effect of PE.
