摘要
首次建立了N-溴代丁二酰亚胺(NBS)-曙红Y(EY)化学发光体系用于检测盐酸二甲双胍,在本化学发光体系中NBS为氧化剂、EY为能量受体.在碱性条件下,NBS氧化盐酸二甲双胍产生稳定的化学发光,向体系中加入十六烷基三甲基溴化胺(CTMAB)后,化学发光显著增强.研究了NBS浓度、EY浓度、CTMAB浓度、NaOH浓度、阀池管长和泵速等因素对化学发光强度的影响,初步探讨了此化学发光可能的机理.结果显示:在最佳实验条件下,盐酸二甲双胍的检测线性范围为2.6×10-8~2.0×10-5g.mL-1,线性回归方程为I=32.763C(×10-7 g.mL-1)+98.16,r=0.999 1,方法检出限(DL=3σn-1/S)为8.5×10-9g.mL-1.对盐酸二甲双胍片剂(标示量为0.25g.片-1)中盐酸二甲双胍定量分析,结果平均为0.2480 g.片-1,相对标准偏差为0.4%,加标回收率为93.8%~102.7%,结果令人满意.
A novel N-bromosuccinimide (NBS) - Eosin Y (EY) chemiluminescence (CL) system for the determi- nation of metformin hydrochloride coupled with flow injection analysis is proposed. It is found that chemilumines- cence is taken by NBS oxidation metformin hydrochlorid with EY as energy transfer in alkaline. This system is en- hanced by hexadecyl trimethyl ammonium bromide (CTMAB). The effects of various factors (NBS concentration, EY concentration, CTMAB concentration, NaOH concentration, peristaltic pump pipe range and flow rates) on the CL system are investigated. The possible mechanism is also studied preliminarily. The line arrange of determination is 2.6×10^-8~2.0×10^-5g·mL^-1 and the equation of linear regression is I=32.763C(×10^-7g·mL^-1)+98.16,r=0.9991, The detection limit (DL=3σn-1/S) is 8.5×10^-9g·mL^-1. Using this CL system, the detection results of metformin hydrochloride in tablets (0. 248 0g/tablet) compared well with the labeled amount (0. 25 g/tablet) , the value of RSD is found to be 0. 4% ,and the recovery are 93.8% - 102.7%.
