摘要
[目的]建立火龙果离体快速繁殖及再生体系。[方法]以火龙果的茎段为外植体,以MS为基本培养基,通过添加不同浓度的6-BA、KT、NAA和IAA,对火龙果进行增殖、伸长和生根培养试验,研究不同激素浓度对火龙果增殖、伸长和生根培养的影响,为火龙果快繁筛选最佳培养基。[结果]不同激素种类和浓度对火龙果的组织培养有不同的影响,MS+6-BA3.00mg/L+IAA0.50mg/L为火龙果的适宜增殖培养基,外植体的增殖数最多,长势旺,而且粗壮;MS+6-BA0.50mg/L+IAA0.50mg/L为火龙果的适宜伸长培养基,芽长4.6cm,生长健壮;1/2MS+IAA1.00mg/L为火龙果的适宜生根培养基,所生根匀称粗壮。[结论]该研究为完善火龙果组织培养快繁技术提供依据。
[Objective] The study aimed to establish the rapid propagation in vitro and regeneration system of Hylocereus undatus. [Method] With stem of Hylocereus undatus as explants, MS basic medium was used as the media by adding different concentration of 6-BA, KT, NAA and IAA, the proliferation, elongation and rooting culture tests were carried out on Hylocereus undatus so as to research the effect of hormone with different concentration on proliferation, elongation and rooting culture of Hylocereus undatus and screen the optimal medium for rapid propagation and regeneration system of Hylocereus undatus. [Result] The hormone with different concentration had different effects on tissue culture of Hylocereus undatus. The optimum proliferation medium for Hylocereus undatus was MS + 6-BA 3.00 mg/L+IAA 0.50 mg/L, on which the explants got more proliferated number, strong growth vigor and sturdiness. The optimum elongation culture medium for Hylocereus undatus was MS+6-BA 0.50 mg/L+IAA 0.50 mg/L, on which the length of bud was 4.6 cm and had strong growth. The optimum rooting culture medium for Hylocereus undatus was 1/2MS+IAA 1.00 mg/L, on which the roots were symmetry and thick. [Conclusion] This study provided the basis for improving the tissue culture propagation techniques of Hylocereus undatus.
