干涉lncRNAs HOTAIR对人乳腺癌细胞MDA-MB-231的影响 被引量:12

摘要 目的:探讨利用RNA干涉沉默HOTAIR基因对人乳腺癌细胞MDA-MB-231的影响。方法:利用脂质体转染MDA-MB-231细胞;qPCR检测HOTAIR基因的表达;检测EMT相关的Snail蛋白水平的表达;流式细胞术检测凋亡率;MTT法检测细胞增殖。结果:干涉HOTAIR基因后细胞中Snail蛋白表达水平下降,细胞凋亡增加。结论:靶向HO-TAIR的序列特异性siRNA可显著抑制HOTAIR基因的表达;干涉HOTAIR基因表达可抑制细胞增殖,促进细胞凋亡。
