摘要
目的:探讨β-榄香烯对肾细胞癌786-0细胞的抗肿瘤作用及机制。方法:浓度为50,100,150,200和250μg/mlβ-榄香烯作用于肾癌786-0细胞24、48和72h,MTT法检测细胞活力。50,150μg/mlβ-榄香烯作用于786-0细胞24h,流式细胞术碘化丙啶(PI)染色检测细胞凋亡和周期分布,免疫印迹法检测PARP裂解和凋亡相关蛋白的表达。结果:β-榄香烯对肾癌786-0细胞的抑制作用随着浓度和时间的增加而增强。流式细胞术结果提示β-榄香烯可诱导细胞凋亡,而对细胞周期分布无明显影响。β-榄香烯可裂解PARP,并下调抗凋亡蛋白Bcl-2和Survivin的表达。结论:β-榄香烯可能通过诱导细胞凋亡发挥对肾癌786-0细胞的抗肿瘤作用。
Objective:To investigate the anti-tumor effect of β-elemene on renal-cell carcinoma(RCC) 786-0 cells and the mechanisms involved.Methods: 786-0 cells were treated with different concentrations of β-elemene for 24,48 or 72 h.Cell viability was determined using MTT assay.Apoptosis and cell cycle distribution were determined with flow cytometry following Propidium iodide(PI) staining.Expression of apoptosis associated proteins was detected by western blot.Results: β-Elemene inhibited the viability of 786-0 cells in a dose-and time-dependent manner.Flow cytometry assay showed that β-elemene induced apoptosis,but had little effect on cell cycle distribution.Western bolt showed that β-elemene cleaved PARP and down-regulated the expression of Bcl-2 and Survivin.Conclusion: The anti-tumor effect of β-elemene on RCC 786-0 cells might be associated with apoptosis induction.
