摘要
目的探讨5-氮杂2-脱氧胞苷(5-AzadC)联合曲古抑菌素A(TSA)对人胃癌SGC-7901细胞生长及Reprimo基因表达的影响。方法 SGC-7901细胞分为对照组、5-Aza-dC、TSA、5-AzadC+TSA组,MTS法测定各组药物对其生长的影响,RT-PCR和Western Blot法分析SGC-7901细胞Rep-rimo基因mRNA和蛋白表达情况。结果 5-AzadC和(或)TSA分别作用24、48、60、72 h后均抑制细胞生长,两药联合组比单药组抑制率显著增强,并且随着药物浓度增加和作用时间延长,抑制率增强更加显著;5-AzadC和(或)TSA干预胃癌细胞72 h后,联合用药组和单药组比较,Reprimo mRNA和蛋白的表达显著增强(P<0.05)。结论 5-AzadC联合TSA干预可抑制SGC-7901细胞生长,抑癌基因Reprimo再表达发挥抑癌作用。
Objective To investigate the effects of 5-Aza-2′-deoxycytidine(5-AzadC) and trichostatin A(TSA) on the expression of Reprimo gene and the growth of human gastric cancer cell line SGC-7901.Methods Experimental cells were divided into four groups by received different treatment: the control group,the group with 5-AzadC;the group with TSA and the group with 5-AzadC and TSA.Cell growth was evaluated by MTS method,expression of Reprimo protein was detected by Western blot and RT-PCR was applied to detect the Reprimo mRNA.Results 5-AzadC and(or) TSA all inhibited the growth of SGC-7901 cells after being treated for 24,48,60,72 h. The inhibitory rate in the union medication group was significantly higher than that in the independent medication group with a time-dependent and concentration-dependent manner.Compared with the independent medication group,the Reprimo mRNA and protein expression of SGC-7901 cells were much more in the union medication group significantly increased independent medication group in 72 h(P〈0.05).Conclusion In the union medication group,the drug is significantly much more effective than that in the independent medication group in inhibiting the proliferation of SGC-7901 cells and significantly up-regulating Reprimo mRNA and protein expression.
