摘要
目的探讨大肠癌中淋巴管密度(LVD)和血管内皮生长因子-A(VEGF-A)的表达与微血管密度(MVD)的关系。方法应用抗podoplanin及抗VEGF-A单克隆抗体检测大肠癌组织及相应正常大肠组织中的LVD,并计数CD34阳性着色的大肠癌组织MVD。结果①大肠癌组织中的LVD明显高于相应的正常组织(P<0.01),LVD与淋巴管内癌栓密切相关(P<0.05)。②VEGF-A在大肠癌组织中的表达显著高于相应正常组织(P<0.01),VEGF-A的表达与MVD(P<0.05)相关但与LVD无关(P>0.05)。③大肠癌中MVD显著高于LVD(P<0.01),但两者无关。结论淋巴管的形成在大肠癌的发生、发展过程中起重要的作用;VEGF-A在大肠癌组织中可诱导微血管形成但与淋巴管的形成无关;大肠癌的MVD显著高于LVD,但两者无相关性,提示血管形成及淋巴管形成在大肠癌的发生、发展过程中可能为相互独立的过程。
Objective To investigate the relationships of lymphatic vessel density(LVD) and the expression of vascular endothelial growth factor-A(VEGF-A) with microvessel density(MVD) in colorectal cancer.Methods Immunohistochemical staining with monoclonal antibodies against human podoplanin and VEGF-A was used to examine LVD and VEGF-A expression in 81 cases of colorectal cancer and corresponding normal tissues and CD34 positively stained MVD was also calculated in the cancer tissue samples.Results The LVD of colorectal cancer samples was significantly higher than that of concordant normal tissues(P0.01) and it was significantly correlated with lymphatic vessel invasion(P0.05).The expression of VEGF-A in colorectal cancer was significantly higher than that in normal tissue(P0.01),VEGF-A was correlated with MVD(P0.05) but not with LVD(P0.05).The MVD in colorectal cancer was significantly higher than the LVD(P0.01),however,no correlation between LVD and MVD was identified.Conclusion Lymphangiogenesis is closely related with the occurrence and progression of colorectal cancer.VEGF-A is assocaited with angiogenesis but not lymphangiogenesis.The MVD in colorectal cancer is significantly higher than the LVD.However,the LVD and MVD are not correlated,suggesting that angiogenesis and lymphangiogenesis may be independent events during colorectal cancer progression.
