五种中药对体外培养的大鼠骨髓间充质干细胞增殖活性的作用 被引量:11

Effects of Five Kinds of Traditional Chinese Medicine on Proliferation Activity in Vitro of Bone Marrow Mesenchymal Stem Cells in Rats

摘要 目的:观察单味中药黄芪、丹参、淫羊藿及复方右归饮药物血清和煎剂以及雪莲花药物血清及其醇提液对体外培养的大鼠骨髓间充质干细胞增殖活性的作用。方法:雪莲花乙醇浸泡后定容,其他单味和复方中药水煎定容,高压灭菌。按10倍于任用剂量进行SD大鼠灌胃,心脏采血,制备血清。取大鼠股骨和肱骨,分离骨髓间充质干细胞,常规培养传代3次后,加入含药血清、煎剂及雪莲花醇提液。药物血清分为15%1、0%5、%和1%浓度,其煎剂及雪莲花醇提液浓度分为10-2、10-3、10-4、10-5,作用3天后用MTT法测定细胞活力。结果:雪莲花含药血清及其醇提液在4个浓度组均显示促进细胞增殖。结论:雪莲花具有促进大鼠骨髓间充质干细胞体外培养增殖活性的作用。 Objective :To investigate the effects of Radix Astragali, Salvia, Herba Epimedii, compound Youguiyin serum and its decoction,Xuelianhua Decoction and its ethanolic extract on proliferative activity in vitro of bone marrow mesenchymal stem cells in rat. Methods :Through high pressure sterilization,after Saussurea lnvolucrate soaked in ethanol and other single herb and compound decoction were administered to SD rats by 10 times of the routine dose to do the preparation of herb serum. The bone marrow mesenchymal stem cells (MSCs) were isolated and cultured from rat femur and humerus by conventional method. The herb serum administered to the P3 MSCs with 15% ,10% ,5% and 1% concentration group. The ethanolic extract of Saussurea In- volucrate and other single herb and compound decoction were administered to the P3 MSCs withl0-2,10-3,10-4,10-5 concentra- tion group. The cells proliferative activity were determined by MTF method after having been cultured 3 days. Results : The herb serum and the ethanolic extract of Saussurea lnvolucrate promoted MSCs cell proliferation with all concentration groups. Conclusion : Saussurea Involucrate showed the positive effect on rat bone marrow mesenchymal stem cells proliferative activity in vitro.
