摘要
目的:检测非小细胞肺癌(NSCLC)中抑癌基因ASPP1、ASPP2的表达,研究其与肺癌临床特征的关系,并探讨其与p53的相关性。方法:利用组织芯片技术结合免疫组织化学Envision法检测ASPP1、ASPP2和p53蛋白在90例NSCLC和25例相应癌旁正常组织中的表达。结果:NSCLC组织中ASPP1的表达显著低于癌旁正常组织(P<0.05),ASPP1的表达与NSCLC的分化程度、临床分期及淋巴结转移相关(P<0.05);ASPP2在NSCLC及癌旁正常组织中的表达差异无显著性(P>0.05),ASPP2蛋白的表达与NSCLC的临床病理特征无关(P>0.05);ASPP1和p53表达呈负相关(r=-0.259,P=0.014),ASPP2和p53的表达状态无关(r=-0.119,P=0.264)。结论:ASPP1蛋白的低表达与NSCLC的恶性进展有关,ASPP1可能在诱导野生型p53细胞凋亡通路中起重要作用,ASPP2蛋白在NSCLC中与p53的表达无关,且可能不具有肿瘤特异性。
Objective To investigate the expression and clinical significance of ASPP1, ASPP2 and p53 and to explore their significance in non-small-cell lung cancer (NSCLC). Methods The expression levels of ASPPl, ASPP2 and p53 protein were examined in lung carcinoma tissue (n = 90), and pericancerous tissues (n = 25)by tissue microarray technique and immunohistochemistry staining. Results The positive rate of ASPP1 was significantly lower in cancer tissues than in their normal adjacent tissues (P 〈 0.05). The expression of ASPP1 had close relation with the differentiation, TNM stage and lymph node metastasis in NSCLC (P 〈 0.05). There was no significant difference between lung cancer tissue and the side-carcinoma tissues in expression of ASPP2 (P 〉 0.05 ). There was a negative correlation between the expressions of ASPP1 and p53 (r = -0.259,P = 0.014), but the expression of ASPP2 was not associated with p53 state (r = -0.119, P = 0.264). Conclusion The decrease of ASPP1 expression may contribute to invasion and metastasis in NSCLC, and ASPP1 may regulate the tumor apoptosis by inducing wide type p53 ; the expression of ASPP2 protein was not associated with the expression of p53 and the tumor specific in NSCLC.
