二甲双胍对晚期糖基化终产物诱导的血管平滑肌细胞增殖的影响 被引量:1

摘要 目的 探讨二甲双胍对晚期糖基化终产物( AGE)诱导的血管平滑肌细胞(VSMC)增殖的影响.方法 采用传代细胞培养法培养VSMC细胞;将100、200、400mg/L的AGE分别作用于VSMC,且设200 mg/L BSA组与空白对照组;10-5、10-3、10-3mol/L的二甲双胍+200 mg/L的AGE分别作用VSMC,于加药后12、24、48及72 h观察增殖变化;后用四氮唑蓝法测定其吸光度值.结果 各浓度AGE组在不同时间均能促进VSMC增殖;其中,100 mg/L AGE组12 h时对VSMC就有增殖作用,而200 mg/L AGE组增殖作用最明显,且在24h作用最强;400mg/L组同200 mg/L组相比无统计学差异(P>0.05);各浓度二甲双胍组在24h、48 h、72 h各组均能抑制VSMC增殖,相比均有统计学意义(P<0.05);各浓度二甲双胍组相比,10-4mol/L、10-3 mol/L组24h抑制作用最强,经比较有统计学意义(P<0.05).结论 二甲双胍对AGE诱导的VSMC增殖有抑制作用. Objective To explore the influence of metformin on adanced glycation end products (AGE) induced by the vascular smooth muscle cells (VSMC) proliferation.Methods The VSMC were cultured by passaged cell cultured method; 100,200,400 mg/L of advanced glycation end products were acting on VSMC respectively,and located 200 mg/L of BSA group and blank control group; 10 -5mol/L,10 -4mol/L,10 -3mol/L of metformin +200 mg/L of AGE were acting on VSMC respectively,and observed the proliferation changes at 12,24,48 and 72 h after dosing the drugs; the absorbance value was determined by MTT method.Results The different concentrations of AGE group can promote proliferation of VSMC at different times; and 100 mg/L of AGE group had proliferation effect to VSMC,at 12 h,200 mg/L of AGE group had the most obvious,and it had the strongest effect at 24 h;there was no statistically significant difference between the 400 mg/L group and the 200 mg/L group ( P 〉 0.05 ) ;the different concentration of metformin at 24 h,48 h,72 h of each group were able to inhibit proliferation of VSMC,and there were statistically significant differences ( P 〈 0.05 ) ; 10 -4 mol/L and 10 -3 mol/L of metformin group had the strongest inhibition compared with the others (P 〈0.05).Conclusion Metformin for VSMC proliferation of AGE induce had inhibited effect.
