摘要
目的探讨槲寄生碱(mistletoe alkali,MA)通过激活Caspase8途径诱导涎腺腺样囊性癌(ACC)细胞凋亡的研究。方法以5-氟尿嘧啶(5-FU)作用组为阳性对照组,以不加药组为阴性对照组,槲寄生碱处理组为实验组作用ACC细胞,采用免疫组织化学染色方法,检测三组细胞中Caspase8蛋白的表达情况。结果 槲寄生碱组Caspase8表达明显高于对照组(P<0.01),并且随着槲寄生碱浓度越高,Caspase8表达越强。结论槲寄生碱可以通过激活Caspase 8途径来诱导ACC细胞凋亡,并呈剂量依赖性。
Objective to study whether alkaloid from mistletoe induces apoptosis of ceils of adenoid cystic carcinoma of salivary gland (ACC cell) via activation of caspase8;methods The ACC cells treated by 5-FU were used as positive control group and the ACC cells without any treatment as negative control group, the ACC cells treated by mistletoe alkali were used as experimental groups, the expression of alkaloid groups is obviously higher than the control groups, Results The expression of caspase8 of alkaloid group was much higher than that of control groups(P〈0.01) ,and the expression is stronger with the higher concentration of alkaloid. Conclusion alkaloid of mistletoe induces apoptosis of ACC cell via activation of caspase8 and the increased easpase8activity is dose dependent.
