外周血生存素的表达与胃癌生物学行为相关性研究 被引量:5

Expression of Survivin in the Peripheral Blood of Patients with Gastric Cancer and Its Relationship with Biological Behaviors of Gastric Cancer

摘要 目的以反转录-聚合酶链式反应-酶联免疫测定(RT-PCR-ELISA)方法,检测胃癌患者外周血中生存素基因的表达。并分析其与胃癌生物学行为的关系,探讨其作为评价胃癌生物学行为及预后检验指标的可能性。方法收集53例胃癌患者外周血,以RT-PCR-ELISA方法检测胃癌患者外周血中生存素mRNA的表达,比较其是否与肿瘤部位、大小、浸润深度、分化程度、淋巴结转移和临床分期具有相关性。结果 53例胃癌外周血中60.4%(32例)生存素mRNA表达阳性。肿瘤>5cm及≤5cm生存素mRNA表达率分别为78.3%、46.0%。浸润深度达浆膜层者外周血生存素mRNA表达率74.2%,高于侵及黏膜下层及肌层的表达率(33.3%),淋巴结有转移者外周血表达率为75.0%,高于无淋巴结转移的表达率(29.4%)。结论生存素mRNA在外周血的表达与肿瘤大小、浸润深度、淋巴结转移和临床分期均有明显相关性,但与肿瘤部位、分化程度无明显相关。外周血生存素的表达可作为评价胃癌生物学行为及预后的参考指标。 Objective Reverse transcriptase polymerase chain reaction and enzyme-linked immuneoassay(RT-PCR- ELISA)was used to assess the expression of survivin mRNA in the peripheral blood of patients with gastric cancer. We investigated it's relationship with biological behaviors of gastric cancer to discuss the possibility of being a method to evaluate the prognosis of gastric cancer. Methods the peripheral blood was collected from 53 patients with gastric cancer. RT-PCR-ELISA was used to assess the expression of survivin mRNA in the peripheral blood of patients with gastric cancer. Calculate the dependability between the expression and the sites, size, infiltration, degree of tumor differentiation,Iymph node metastasis and clinical stages. Results The expression rate of survivin mRNA in the peripheral blood of the 53 cases of patients with gastric cancer is 60.4%(32 cases). The expression rate for the tumor size 〉5 cm and ≤ 45 cm is according to 78. 3% .46.0% . The expression rate of patients whose plasma membrane were infiltrated is 74.2% ,which is higher than others(33.3%), when there was lymph node metastasis, the expression rate is 75.0%, which is higher than the rate when there was no lymph node metastasis(29.4%). Conclusion The expression of survivin in the peripheral blood was related with the size, infiltration, lymph node metastasis and clinical stages, but was not correlated with sites or the degree of tumor differentiation. The expression of survivin in the peripheral blood might be a referenced indicator in evaluating the biological behaviors and prognosis of gastric cancer.
