摘要
目的检测氯通道ClC-2在人非小细胞肺癌中的表达情况,探讨ClC-2在非小细胞肺癌中表达的临床意义,为ClC-2成为临床肺癌治疗的新基因靶点提供新思路。方法实验分为对照组和肺良性病组、肺鳞癌组和肺腺癌组,应用半定量RT-PCR及Western blot方法检测各组ClC-2mRNA及ClC-2蛋白表达水平并对ClC-2在非小细胞肺癌中的表达情况进行初步分析。结果 ClC-2mRNA表达水平上与对照组相比,肺鳞癌组和肺腺癌组中的ClC-2mRNA表达量明显增多,但肺良性病与正常肺组织之间无显著差异。ClC-2蛋白表达量在非小细胞肺癌及肺良性病组均比正常肺组织表达增多。结论两种实验方法均证实了ClC-2在非小细胞肺癌中明显表达。提示ClC-2通道可能在临床肺癌侵袭、发展及肺部疾病发生过程中起重要作用,ClC-2可能成为临床治疗非小细胞肺癌的新分子靶点,深入研究ClC-2与肺癌发生发展关系中提供病理生理基础。
Objective Detect expression situation the chloride channel-clc-2 in the nonsmall-eell lung cancer,discuss clinic significance of the ClC-2 expression in the nonsmall-cell lung cancer. To provide the new idea for-ClC-2 being new gene target spot of the clinic lung cancer therapy. Methods The experiment is divided to the control group and benign lung disease group,apply the method of semiquantitative RT-PCR and Westernblot to detect each group's the level of the protein expression of the ClC-2mRNA and ClC-2, and to proceed preliminary analysis for ClC-2 expression situation in the nonsmall-cell lung cancer. Results To compared with control group in the level of the ClC-2mRNA expression, the expression number of the ClC-2mRNA is obviously increase in the group of the lung squamous carcinoma and lung adenoc,arcinoma, however, there is no observe differences between benign lung disease and normal lung tissue. The protein expression of ClC-2 in the group of nonsmall-cell lung cancer and benign lung disease both increase compare with the normal lung tissue. Conclusion Two experimental methods both confirm that ClC-2 expresses obviously in the nonsmall-cell lung cancer. It gives a hint that channel-ClC-2 may play a important role in the affect of clinical lung cancer, development and the occurrence process of the lung cancer. ClC-2 may become the new molecular target for clinical therapy in the nonsmall-cell lung cancer and penetrate to study to provide the pathophysiological base between the ClC 2 and lung cancer's occurrence and development relation.
