克拉霉素经NF-κB途径抑制肺炎支原体抗原诱导A549细胞产生IL-8 被引量:2

Effect of Clarithromycin on IL-8 Production in A549 Cells Induced by Mycoplasma pneumoniae Antigen

摘要 目的观察克拉霉素对肺炎支原体抗原诱导A549细胞产生IL-8的影响,为临床治疗肺炎支原体肺炎提供理论依据。方法体外培养肺炎支原体并提取其抗原,用不同浓度抗原刺激预先孵育过克拉霉素的A549细胞,ELISA检测IL-8的分泌情况;同时提取处理前后A549细胞的核蛋白,Western blot检测核蛋白中p65水平的变化。结果肺炎支原体抗原能诱导A549细胞产生IL-8,0.1~10μg/ml克拉霉素对其具有一定的抑制作用。此外,克拉霉素也能明显降低处理后A549内核蛋白的p65水平。结论克拉霉素可抑制肺炎支原体抗原诱导的A549细胞分泌IL-8,其机制可能与抑制NF-κB的激活有关。 Objective To investigate the effect of clarithromycin on IL- 8 production in A549 cells induced by Mycoplas- rna pneumoniae antigen. Methods M. pneumoniae was cultured in vitro and its antigen was extracted. Then the antigen was added to the A549 cells pre - incubated with different concentrations of clarithromycin. The level of IL - 8 was detected by ELISA. At the same time, nucleoprotein was obtained and the p65 level was detected by Western blot. Results M.pneumoniae antigen could induce IL - 8 production in A549 cells. The content of IL - 8 was decreased after precultured with 0.1-- 10μg/ml of clarithromycin. In addition, the p65 level in nucleoprotein was also attenuated by clarithromycin. Conclusions Clarithromycin can inhibit the production of IL- 8 induced by Mycoplasrna pneumoniae antigen, which may be mediated by the inhibition of NF-κBactivation.
