AKT2在乳腺癌中的表达及其与微血管密度的相关性研究

摘要 目的:探讨AKT2在乳腺癌和正常乳腺组织中的表达及其微血管密度变化,分析其表达水平与临床病理特征的关系。方法:用免疫组化SP法检测92例乳腺癌组织、35例正常乳腺组织中AKT2的表达及微血管密度。结果:乳腺癌、正常乳腺组织AKT2阳性表达率分别是60%(55/92)和11.43%(4/35),乳腺癌组织AKT2阳性表达率明显高于正常乳腺组织(P<0.05)。AKT2的阳性表达与乳腺癌的组织学分级、肿瘤大小、ER、PR表达无关(P>0.05),同淋巴结转移和TNM分期、CerbB-2差异有统计学意义(P<0.05)。乳腺癌组织MVD明显高于正常乳腺组织MVD:(36.94±1.60vs6.74±2.87,P<0.05)。MVD值与乳腺癌的组织学分级、ER、PR表达无关(P>0.05),在肿瘤大小、淋巴结转移和TNM分期、CerbB-2表达差异有统计学意义(P<0.05)。AKT2阳性表达的乳腺癌MVD为:40.87±10.97,阴性表达MVD为:30.25±9.49,两者之间的差别有统计学意义(P<0.05)。结论:AKT2高表达与微血管生成参与乳腺癌的发生、发展和浸润转移,可作为判断乳腺癌生物学行为的重要指标。 Objective: To investigate the expressions of AKT2 and changes of microvessel density(MVD) in breast cancer and normal breast tissues, and analyze their relationship with tumor clinicopathological features. Methods: The expression of AKT2 and MVD were detected with the immunohistochemical method in 92 cases of breast cancer and 35 cases of normal breast tissues~ Results: The positive rate of AKT2 was significantly higher in breast cancer than that in normal breast tissues(60% vs 11.43%,P〈0.05). In breast cancer, the positive rate of AKT2 protein was significantly associated with lymph node metastasis, TNM stage and CerbB-2 expression(P〈0.05),but were not with the histological grade, tumor size, ER and PR expression(P〉0.05). The MVD significantly higher in breast carcinoma than that in normal breast tissues(36.94 ± 11.60 vs 6.74 ± 2.87,P〈0.05) in which MVD was significantly correlated with tumor size, lymph node metastasis, TNM stage and CerbB-2 expression(P〈0.05), but not with the histological grade, ER and PR expression (P〉0.05). The MVD was significantly higher in breast cancer with positive AKT2 expression than in that with negative AKT2 expression(40.87±10.97vs 30.25±9.49,P〈0.05). Conclusion: The higher expression of AKT2 and angiogenesis is involved in the incidence,development, invasion and metastasis of breast cancer, which may be used as an important indications of biological behavior of breast cancer.
