摘要
以放线菌WS-22011为研究对象,通过研究摇瓶发酵和发酵罐发酵两种不同发酵方式下WS-22011产生的次生代谢产物多样性及产量随时间变化的动态趋势,确定WS-22011最佳发酵方式及兼顾代谢产物多样性、产量的最佳发酵时间。结果表明,对于WS-22011菌株来说,在发酵罐发酵和摇瓶发酵两种发酵方式下产生的次生代谢产物及多样性基本一致,两种发酵方式均可以保证次生代谢产物的多样性和产量,但次生代谢产物最大总产量出现的时间有所差异,从节约时间及成本考虑,可首选发酵罐发酵;而如果需得到所有的次生代谢产物,则首选摇瓶发酵,且发酵时间需适当延长。通过制备分离得到了4个具有较高活性的化合物,LC-MS分析结果表明为碰撞霉素。
The dynamics of diversity and yield of secondary metabolites according to fermentation time of Actinomycete WS-22011 under shake-flask fermentation and tank fermentation was studied to optimize the fermentation method and time in consideration of both diversity and yield of secondary metabolites.The results showed that the diversity of WS-22011's secondary metabolites were basically similar under both fermentation methods,but the time for maximum metabolic production differed.Therefore,tank fermentation would be a better choice for saving time and cost;While shake-flask fermentation with long fermentation time should be used if the diversity of secondary metabolites was in first consideration.4 active compounds were isolated,purred and identified as collismycins by LC-MS.
